MULTIPLE SEQUENCE ALIGNMENT

For the Multiple Sequence Alignment, Clustal Omega was used (with default settings) in combination with Boxshade (output format RTF_new) , for shading the conserved regions.

 

The first MSA carried out included all human Tau isoforms, 9 in total. One gene encodes all isoforms, and any differences between are due to alternative splicing. Therefore, as expected, the main difference observed between the isoforms is length. Another MSA was carried out, using the human PNS isoform, three mouse (Mus musculus) isoforms and two monkey (Macaca mulatta) isoforms. This was done to determine the level of conservation between different species. Tau shows high conservation between species, especially in the C-terminus, where the tubulin binding repeats are located.
